The Earth-Minbari war began in 2245 when an Earthforce taskforce, seeking out the Minbari, and led by the EAS Prometheus (Captain Jankowski commanding), opened fire on a Minbari convoy en route to Z'Ha'Dum. Among the Minbari killed in the assault was Delenn's mentor, and Grey Council leader, Dukhat.

With the death of her mentor, Delenn, then a candidate member of the Grey Council, declared vengeance upon those who had killed their iconic leader. Her vote broke a tie within the Grey Council, and began Earth's bloodiest conflict in history.

Within three years, in 2248, the Minbari had reached Earth, where they prepared for the final annihilation of the Human species. Already war weary, and looking for a way to spare humanity, Delenn ordered that a few humans should be brought aboard the Grey Council's flagship for study. One, Jeffrey Sinclair, was then interrogated, tortured, and then mindwiped.

During this study, using their vaunted "triluminary," the Minbari came to believe that Humans and Minbari shared souls. Because, as a law of Valen, "Minbari do not kill Minbari", they felt they could no longer kill humans, and therefore contrived their surrender to the humans, and ended the war.
